HLA Transgenic Mice
EpiVax maintains human leukocyte antigen (HLA) DR3 transgenic mouse colonies and has access to DR4 transgenic mouse colonies to assess immunogenicity of vaccines and protein therapeutics in the context of a human-like antigen-presentation system.
HLA transgenic mouse colonies are maintained at Taconic Farms and where these mice are housed in pathogen-free isolators. Sentinel mice are tested regularly for a panel of murine bacterial and viral infectious agents. Transgenic mice are genotyped by standard PCR using genomic DNA extracted from tail tissue and primer pairs specific for HLA transgenes. HLA expression levels are assessed by flow cytometry using mouse peripheral blood mononuclear cells immunostained for HLA. Available assay formats include:
- Serum antibody levels by ELISA,
- T cell cytokine expression (ELISpot and multiplex ELISA)
- T cell phenotype using flow cytometry and cytotoxic T cell activity
